This online course includes all the recorded presentations, course materials, case studies, and tests from the live course held on May 16-19, 2019. CME/CMLE or SAMs is available with the purchase of this course until December 31, 2020.


This course features an intense update and review of molecular diagnostics for those prepping for the Molecular Genetic Pathology (MGP) Subspecialty Certification Exam as well as other certification exams, those who need a refresher, or anyone looking to add molecular testing to their current pathology practice. Participants have the opportunity to learn directly from recognized experts in the field of molecular pathology. Learn at your own pace and rewatch lectures as many times as you'd like.

Collapse Circulating Tumor DNA Testing: Advances, Challenges, and Applications (+AMP Certificate)

This AMP certificate program contains 5 hours of lecture content, the associated pdf slides of the lectures, a post-course exam, and an AMP Certificate of Completion. It does not include CME/CMLE or SAM.


Circulating tumor DNA has shown much promise as a minimally invasive test that could allow for disease detection, identification of targetable variants, and monitoring of tumor response. This AMP certificate program, featuring expert speakers in the field, will: define circulating tumor DNA, explain technical challenges, describe approaches for assay design and validation, highlight clinical applications of testing, and will review selected new published studies and guidelines. Participants can expect, upon completion of this certificate program, to be familiar with the current state of circulating tumor DNA testing for personalized medicine.

    The advent of targeted therapies based on predictive biomarkers has dramatically altered the role of the molecular pathologist in lung cancer patient care. This webinar reviews the new recommendations published in the revised 2018 CAP/IASLC/AMP lung cancer predictive biomarker guidelines (Lindeman et al., J Mol Diagn, 2018). This webinar, reviewing the evidence and logic behind the new recommendations, is designed for molecular pathologists, pathologists, oncologists, clinicians, trainees, and multidisciplinary tumor boards to help improve NSCLC patient care. Topics to be covered include recommendations regarding new predictive biomarkers; recommendations for testing for EGFR and ALK TKI resistance; and recommendations regarding new testing methodologies including the use of multiplexed genetic sequencing panels (e.g., NGS testing) to identify other treatment options beyond EGFR, ALK, and ROS1 as well as cell-free circulating DNA testing for EGFR resistance mutations. The revised 2018 CAP/IASLC/AMP lung cancer predictive biomarker guidelines will impact the daily practice of molecular pathologists and clinicians in the care of nearly 150,000 eligible lung cancer patients each year, regardless of whether in a community practice, a large reference lab, or an academic setting, and regardless of their level of expertise or experience.

    In this webinar, Dr. Karen Weck will present the recommendations of The AMP Pharmacogenomics Working Group of the Clinical Practice Committee for inclusion of variants in CYP2C19 genotyping panels. The working group has published consensus, evidence-based recommendations using criteria such as allele function, population frequency, and availability of reference materials. The recommendations include a Tier 1 minimum set of alleles and defining variants that should be included in all clinical CYP2C19 PGx tests, and a Tier 2 list of additional optional alleles that do not currently meet all of the criteria for inclusion in Tier 1. These recommendations are intended to aid clinical laboratory professionals when designing and validating CYP2C19 genotyping assays, promote standardization of testing across different laboratories, and complement existing clinical guidelines for pharmacogenetic testing.

    In oncology, there are a variety of potential clinical applications for a minimally invasive blood test for DNA released from tumor cells. Such applications include detecting early stage disease, tracking response to therapy, prompt identification of disease relapse or recurrence, and recognition of clonal evolution.  Numerous small-scale studies have demonstrated that quantitation of mutations in circulating tumor DNA (ctDNA) is feasible from the blood of cancer patients. The increased adoption of targeted cancer therapies underscores the need to augment invasive tissue biopsies with alternative detection methods that rapidly monitor treatment response. Although most studies have been proof of principle, ctDNA methods hold great promise in cancer diagnostics.

    Description: This presentation will introduce the audience to pathways of DNA repair in human cells, in particular, the homologous recombination (HR) pathway which responds to and repairs DNA double-strand breaks (DSBs) during DNA replication. The BRCA1 and BRCA2 genes code for proteins that are key players in the HR pathway and we will discuss their mechanistic roles in HR. We will discuss germ line mutations in the BRCA genes and how to differentiate a pathogenic mutation from a benign polymorphism as well as variants of uncertain significance and the implications for future cancer risk. We will introduce the concept of loss-of-heterozygosity of the wild type BRCA allele in tumors from patients who are germ line carriers of BRCA mutations. We will introduce the idea of synthetic lethality and how PARP inhibitors (PARPi) were discovered to selectively kill BRCA (HR deficient) tumors. Finally, we will speculate on the mechanism of action of PARPi and mechanisms of resistance in both cell lines and patients who have relapsed during PARPi treatment.
