Speakers presented a variety of current reference material development projects for molecular testing of inherited and acquired somatic genetic disorders followed by group discussion.

This course is designed for pathologists, residents, laboratory directors and technologists who have some prior exposure to molecular diagnostic testing. The course features introductory information about the state of the science as well as a series of case studies illustrating some of the most current principles and practices in the field of Molecular Pathology.

    Laboratory diagnostics of HIV continue to evolve with antigen/antibody and molecular methods becoming increasingly more sensitive, specific, and rapid. Moreover, laboratory implementation of the CDC/APHL testing algorithm incorporating these assays may result in improved diagnosis of acute HIV but may also be challenging to interpret. The overall goal of this webinar is to discuss diagnostic methods for HIV and the current testing algorithm through the presentation of actual clinical cases. The webinar will conclude with a brief discussion of emerging or novel diagnostic methods and their impact on HIV diagnosis.

    Collapse Next-Generation Sequencing (NGS) Technologies for HIV Drug Resistance Testing

    HIV-1 exists within individuals as a complex mixture of variants often referred to as a quasispecies. During the past 10 years, there have been many studies of the distribution and clinical significance of "low-abundance" or "minority" drug-resistance variants present in plasma in proportions below 20% to 30% -- the limit of detection of standard genotypic resistance testing using dideoxy-terminator "Sanger" sequencing. HIV-1 "deep" sequencing using NGS technologies have been widely used in research settings to study the clinical significance of low-abundance drug resistance mutations. In addition, NGS is commercially available for several niche clinical applications such as determining HIV-1 tropism before using a CCR5 inhibitor.
    In this webinar, Dr. Shafer will review the scenarios in which low-abundance HIV-1 drug-resistance mutations are most clinically relevant, the most influential studies of NGS for HIV-1 drug resistance testing, and the practical aspects of NGS that have so far slowed its widespread adoption for most routine HIV-1 genotypic resistance testing.

    Zika in Latin America: Clinical Aspects by Dr. Cecilia Perret

    Dr. Perret reviews the epidemiology of the Zika outbreak in Latin America, some clinical aspects, evidence of a Zika infection, and its complications (congenital disease and Guillain Barre Syndrome). In addition, she will discuss the local response to a series of travelers infected with Zika, including a pregnant woman and a sexually transmitted infection.

    Zika Virus Diagnostics by Dr. Benjamin Pinsky

    Zika virus (ZIKV) is an Aedes mosquito-borne flavivirus that emerged in Brazil in 2015 and then rapidly spread throughout the tropical and subtropical Americas. Based on clinical criteria alone, ZIKV cannot be reliably distinguished from infections with other pathogens that cause an undifferentiated systemic febrile illness, including infections with two common arboviruses, dengue virus and chikungunya virus. This presentation details the methods that are available to diagnose ZIKV infection.
