CHIPping Away at the Mysteries of Hematopoiesis: The Science and Clinical Implications of Clonal Hematopoiesis of Indeterminate Potential

The concept of a pre-cancerť clonal proliferation is fundamental to our understanding of the natural evolution of a variety of cancer types and an active area of research. Recent recognition of pre-malignant clonal hematopoiesis has been yet another exciting discovery in this area. The speakers will discuss their work in identifying Clonal Hematopoiesis of Indeterminate Potential (CHIP) and its relationship to both aging and the subsequent development of recognized hematopoietic malignancies.

- Explain the concept of clonal hematopoiesis.

- Describe how clonal hematopoiesis can be evaluated using markers identified by high-through sequencing.

- Summarize current knowledge regarding the association between clonal hematopoiesis and the development of recognized hematopoietic cancers, particularly myelodysplastic syndromes.

Description:

Since the publication of the World Health Organization (WHO) Classification of Tumors of Hematopoietic and Lymphoid Tissues in 2008, there have been significant advances in our knowledge of the biology and genetics of hematopoietic malignancies resulting in improved diagnostic and prognostic criteria, disease classification and therapeutic strategies. The 2016 WHO Classification of Tumors of Hematopoietic and Lymphoid Tissues incorporates these advances with revisions to current entities and addition of provisional entities. This AMP certificate program will cover the major changes made in the 2016 WHO revision. Participants will learn from experts in the field covering the following topics: B lymphoid neoplasms, Peripheral T cell lymphomas, Acute Leukemias, Myeloproliferative Neoplasms, Myelodysplastic syndromes and Myelodysplastic/myeloproliferative neoplasms. An update on the clinicopathologic features and classification of hematologic malignancies will be presented providing participants with skills and knowledge to accurately diagnose and subclassify hematologic malignances according to the revised 2016 WHO criteria.

    Collapse 2017 AMPlicons: A Practical Molecular Toolkit and Case Studies Online Course

    This course is designed for pathologists, residents, laboratory directors and technologists who have some prior exposure to molecular diagnostic testing. The course features introductory information about the state of the science as well as a series of case studies illustrating some of the most current principles and practices in the field of Molecular Pathology. Attendees from all career levels will benefit from this staple offering.

     

    The objective of this course is to increase basic and applied pathology knowledge, focusing on the molecular diagnosis of disease and to facilitate knowledge acquisition regarding issues and challenges related to care and prevention. This course is designed to meet the participants' educational needs in the physician competency area of Medical Knowledge, as defined by the Accreditation Council for Graduate Medical Education (ACGME) and the American Board of Medical Specialties (ABMS), and to support participants' lifelong learning towards a goal of promoting patient safety and improving patient care.

    Contemporary hot topics in the clinical molecular diagnostics laboratory will be introduced, followed by an overview of genomic testing to build the molecular diagnostic "toolkit." Short case presentations will demonstrate practical applications of a wide range of methodologies ranging from common commercial platforms to traditional amplification methods.

    This online course includes all the recorded presentations, course materials, and case studies from the live 2018 Molecular Pathology Outreach Course held on October 31, 2018 in San Antonio, TX.

    This course is designed to provide an overview of techniques and instrumentation used from the framework of the "Molecular Pathology/Molecular Diagnostic Toolkit." In this course, faculty members will present case studies to demonstrate the toolkit’s methodologies and technologies. Emphasis will be placed on molecular testing in the areas of clinical genetics, solid tumors, hematopathology, and infectious diseases. The application of novel technologies, such as next generation sequencing and informatics, will also be highlighted.

    Welcome to the Fellowship-in-Service-Examination brought to you by the Molecular Genetic Pathology (MGP) Program Directors and the Association for Molecular Pathology.

    These exam questions were written by MGP faculty from many MGP-Fellowship institutions and cover the range of topics listed below. For each examination, a total of 45 questions populated from a question bank of approximately 200 questions will be randomly generated. At the end of the examination, you will receive your results along with the correct responses, an explanation of the correct choice, and references for further information. You will only receive this information for each question answered, so you are encouraged to answer all questions, regardless of whether or not you know the correct answer.

    You are welcome to take the exam as many times as you like, but please note that participants will be charged a small administrative fee ($25 for AMP members, $50 for non-members) each time they take the exam.

    This presentation summarizes the findings of the Association for Molecular Pathology Chronic Myeloid Neoplasm Working Group. Chronic myeloid neoplasms (CMNs) are defined as a complex group of hematopoietic disorders, encompassing myelodysplastic syndromes (MDS), myeloproliferative neoplasms (MPNs), the overlap entities (MDS/MPNs), and systemic mastocytosis. The goal of this group was to distill the vast amounts of literature on the mutational profiles of the CMNs into high impact information that would aid molecular pathologists in the development of next generation sequencing (NGS) myeloid panels. This work identifies 34 genes as the minimum recommended testing list and provides information on the frequency of these genes in the various CMNs as well as their prognostic and therapeutic import. In addition, the findings highlight the recurrent patterns of mutational clonal evolution in these patients, uncovering critical insight into the biology of these neoplasms.

    Following a brief overview on the difficulties encountered in the definition of the epidemiology, classification, diagnosis, prognosis, and therapy of peripheral T/NK-cell lymphomas, nodal peripheral T-cell lymphomas (PTCLs) will be discussed in detail. These neoplasms account for more than 60% of tumours stemming from peripheral T-lymphocytes. In the Revised WHO Classification of Tumours of Haematopoietic and Lymphoid Tissues, they correspond to 4 entities: PTCL, not otherwise specified (NOS), PTCLs related to follicular helper T-lymphocytes (FTH), ALK+ anaplastic large cell lymphoma (ALCL), and ALK- ALCL. Tumour related to TFH cells represent a new category, including angioimmunoblastic T-cell lymphoma (AITL), follicular T-cell lymphoma and neoplasms lacking the morphologic characteristics of the previous two subtypes but also stemming from TFH-elements. The latter group was in the past included among PTCLs/NOS. ALK- ALCL was promoted from provisional to accepted entity because of emerging molecular evidences. In addition, the new provisional category of breast implant-associated ALCL was included. Most of the changes introduced in the Revised WHO Classification by comparison with the 2008 fourth edition are due to the extensive application of molecular techniques, including gene expression profiling and next generation sequencing. All the changes will be highlighted and discussed.
